Tango Therapeutics Commences TNG462 Phase 1/2 Trial, Administering Initial Dose to Patients Affected by MTAP-deleted Solid Tumors

Tango Therapeutics, Inc. (NASDAQ: TNGX) has recently initiated a phase 1/2 clinical trial for their drug candidate, TNG462, targeting patients with MTAP-deleted solid tumors. TNG462 is a PRMT5 inhibitor that works in conjunction with MTA and has received Fast Track designation from the U.S. Food and Drug Administration.

MTAP deletions are found in approximately 10-15% of solid tumors, including those with limited treatment options. Currently, there are no FDA-approved treatments specifically designed for cancers with MTAP deletion. Dosing the first patient with TNG462 demonstrates Tango Therapeutics' dedication to developing effective therapies for patients with MTAP-deleted cancers. Barbara Weber, M.D., President and CEO of Tango Therapeutics, highlights the potential impact of their drug candidate, noting that TNG462, along with their brain-penetrant PRMT5 inhibitor, TNG908, which is being evaluated in a separate phase 1/2 study, has the potential to address a wider range of indications.

Both TNG462 and TNG908 are potent PRMT5 inhibitors that selectively target cancer cells with MTAP deletion. MTAP deletion leads to the accumulation of MTA, an inhibitory PRMT5 co-factor. TNG462 binds to and inhibits PRMT5 in the presence of MTA, effectively killing cancer cells with MTAP deletion while sparing normal cells. Preclinical studies have demonstrated significant tumor regression in various cancer models with the use of both TNG462 and TNG908.
