Regeneron and Parabilis Medicines Partner to Develop Novel Antibody-Helicon Therapies

Regeneron Pharmaceuticals has entered into a strategic research collaboration with Parabilis Medicines to develop multiple therapeutic candidates based on Parabilis’ Helicon™ peptide platform, with a focus on Antibody-Helicon™ Conjugates (AHCs).

The collaboration aims to develop a new class of therapies designed to target difficult and historically “undruggable” intracellular proteins across several therapeutic areas.

Helicons are stabilised alpha-helical peptides that can enter cells and interact with intracellular protein targets, including flat protein surfaces that are difficult for traditional small molecules to bind to. The companies will explore the use of Helicons both as stand-alone treatments and in combination with antibodies through AHC technology.

The partnership will combine Parabilis’ Helicon platform with Regeneron’s VelocImmune® antibody technology. The approach is intended to use antibodies to deliver Helicon payloads directly into target cells, allowing selective modulation of intracellular proteins.

Under the agreement, Parabilis will receive $125 million from Regeneron, including a $50 million upfront payment and a commitment by Regeneron to invest $75 million in Parabilis’ next equity financing, subject to certain conditions.

Parabilis is also eligible to receive development, regulatory and commercial milestone payments, along with tiered royalties on future net sales of approved products arising from the collaboration. With five initial targets included in the agreement, the total potential milestone value could reach approximately $2.2 billion. Additional targets may also be added through further option payments from Regeneron.

Under the terms of the collaboration, the companies will jointly discover new Helicons and AHCs, while Regeneron will lead development, manufacturing and worldwide commercialisation activities.
