NTx Bio Supports Baylor College of Medicine in Manufacturing Personalised Cancer Vaccine for Phase 1 Melanoma Trial

NTx Bio is supporting Baylor College of Medicine in the manufacturing of a personalised cancer vaccine being evaluated in Diakonos Oncology’s Phase 1 trial for patients with refractory melanoma.

Baylor researchers are using NTx Bio’s NTxscribe platform to amplify tumour mRNA from small surgical biopsy samples. The process is an important part of the double-loaded dendritic cell vaccine platform used to develop Diakonos Oncology’s DOC1021 (dubodencel).

NTxscribe is designed to preserve the full-length range of the patient’s tumour transcriptome during mRNA amplification. This helps retain a broad representation of tumour antigens for use in the personalised vaccine.

The Phase 1 trial is evaluating DOC1021 in patients with refractory melanoma whose disease has stopped responding to standard treatments, including immune checkpoint inhibitors. The therapy is designed to use the patient’s own immune cells to generate a targeted response against the tumour.

NTxscribe supports the manufacturing process by automating mRNA generation, reducing manual handling and helping improve consistency between batches. The platform uses a continuous-flow hollow fibre bioreactor architecture and is designed to provide purified mRNA with limited manual intervention.

The NTxscribe FLEX system also uses single-use, closed cassettes to simplify changeovers between patient samples and support increased production volumes. Its scalable design is intended to support early-stage clinical programmes while providing a pathway towards higher-throughput manufacturing.

The collaboration demonstrates the use of automated mRNA production technology in personalised cancer vaccine manufacturing and supports efforts to streamline the development of patient-specific therapies.
