New England Biolabs Launches Low-Bias Small RNA Library Prep Kit to Improve RNA Diversity Analysis

New England Biolabs (NEB®) has introduced the NEBNext® Low-bias Small RNA Library Prep Kit, a new solution designed to reduce bias in small RNA sequencing and better reflect the true diversity of RNA species in research samples. This latest offering builds on more than a decade of internal research and development aimed at addressing limitations in traditional small RNA library preparation methods.
The kit features a novel splint adaptor that enhances ligation efficiency and sensitivity, improving the representation of small RNA molecules—including rare and non-coding types—during sequencing. It also offers a faster protocol, completing the workflow in approximately 3.5 hours, and supports a wider range of input amounts. Standard and 2′-O-methylated samples can be processed using the same streamlined protocol.
Additional features include compatibility with up to 480 unique dual index (UDI) primer pairs, which allow for flexible multiplexing. The kit is designed with a shelf life of up to 18 months, offering both reliability and long-term usability.
This launch follows NEB's long-standing work in reducing bias in RNA sequencing. Earlier studies, including those published in 2012 and 2020, identified and addressed inefficiencies in adaptor ligation—one of the main sources of sequencing bias. Building on these findings, the current kit introduces further protocol enhancements that improve accuracy and performance beyond earlier versions.
By minimising technical variation, the NEBNext Low-bias Small RNA Library Prep Kit aims to support more accurate and comprehensive RNA studies, helping researchers explore the full range of small RNA molecules in health, disease, and development.
