Innate Pharma Completes Phase 1 Dose Escalation Enrolment for IPH4502 ADC Study

Innate Pharma has completed patient enrolment in the dose-escalation stage of its Phase 1 IPH4502-101 clinical trial, which is evaluating IPH4502, the company's proprietary Nectin-4-targeting exatecan antibody-drug conjugate (ADC), in patients with advanced solid tumours.
The open-label, multi-centre study is assessing the safety, tolerability and preliminary anti-tumour activity of IPH4502 as a single treatment in patients with Nectin-4-positive advanced solid tumours. These include urothelial carcinoma, non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), as well as breast, ovarian, gastric, oesophageal and colorectal cancers.
A total of 76 patients have been enrolled in the dose-escalation stage across clinical sites in France and the United States. Preliminary results are expected by the end of the year and will be used to determine the optimal dose for the next stage of Phase 1 development in selected tumour types.
According to the company, IPH4502 has continued to demonstrate a favourable safety profile, with limited haematological toxicity observed so far. Early clinical findings have also shown preliminary anti-tumour activity in heavily pre-treated patients, including objective responses in urothelial carcinoma following enfortumab vedotin treatment, as well as in patients with NSCLC and HNSCC.
IPH4502 is an investigational Nectin-4-targeting ADC that combines a humanised anti-Nectin-4 antibody with the topoisomerase I inhibitor exatecan through a proprietary stable linker. The linker is designed to slow the release of exatecan into the bloodstream, with the aim of reducing toxicity. Preclinical studies have demonstrated anti-tumour activity in models resistant to enfortumab vedotin and in tumours with low or varied Nectin-4 expression, supporting its potential use across multiple solid tumour types.
The IPH4502-101 Phase 1 study is ongoing in patients with advanced solid tumours known to express Nectin-4.
