EuMentis Therapeutics Receives FDA Approval to Begin Phase 2 Trial of EM-221 for Schizophrenia

EuMentis Therapeutics, a clinical-stage pharmaceutical company developing treatments for schizophrenia and other central nervous system (CNS) disorders, has received clearance from the U.S. Food and Drug Administration (FDA) to begin a Phase 2 clinical trial of its investigational therapy, EM-221.

EM-221 is a novel inhibitor of the enzyme phosphodiesterase 10A (PDE10A), which plays a role in regulating dopamine and glutamate signalling in brain regions associated with schizophrenia. Unlike existing antipsychotic drugs, which typically work by blocking dopamine receptors and may cause side effects such as sedation, movement disorders, and weight gain, EM-221 targets a different mechanism. It aims to modulate brain activity in a more precise way, potentially reducing side effects while improving treatment outcomes.

The FDA’s authorisation of the Investigational New Drug (IND) application allows EuMentis to advance EM-221 into Phase 2 development. The company is planning to launch the trial later this year, focusing on evaluating the drug's safety, effectiveness, and tolerability in patients with schizophrenia.

EM-221 is designed as a next-generation therapy to address both the positive symptoms (such as hallucinations and delusions) and negative symptoms (including social withdrawal and lack of motivation) of schizophrenia. Preclinical and Phase 1 studies have shown promising results, indicating a broader therapeutic window and better tolerability compared to currently available treatments.

Schizophrenia affects nearly four million people in the United States alone, and there remains a significant need for new treatments that are both effective and easier for patients to tolerate. EuMentis believes that EM-221 may offer a meaningful alternative to traditional antipsychotic therapies, which many patients find difficult to continue long-term due to side effects.

By focusing on the PDE10A pathway, EM-221 represents a novel approach in psychiatric medicine and may offer a new treatment option for individuals living with this challenging and chronic condition.
