Cell Line Development: Multi-Transposase Strategies for Complex Molecular Assembly and Modulated Expression

While transposase-mediated gene insertion has revolutionized high-producing cell line development (CLD), relying on a single system limits engineering potential. By deploying multiple orthogonal Leap-In transposases® with non-overlapping integration sites, we have expanded the CLD toolkit far beyond standard biologics expression.

Drawing from recent publications and case studies, this talk demonstrates how orthogonal multi-transposase strategies maximize complex molecular assembly and advanced cell line customization. We will highlight practical approaches to fine-tuning post-translational modifications—specifically modulating glycan profiles and fucosylation—and seamlessly integrating multi-domain assemblies into stable clonal lines to address tough next-generation therapeutic challenges.

Gavin Barnard

Gavin joined ATUM as Chief Scientific Officer earlier this year. Previously, he held scientific leadership roles at Genentech/Roche, Eli Lilly, Merck, Adimab, and GlycoFi. Gavin earned his Ph.D. in Bioengineering from Dartmouth College in Dr. Tillman Gerngross's lab and holds a Bachelor's degree in Chemical Engineering from the University of Pretoria.
